Report: Missing NYC woman found dead in Turkey

ISTANBUL (AP) ? A New York City woman who went missing while vacationing alone in Istanbul was found dead on Saturday and police detained nine people for questioning in connection with her case, Turkey's state-run news agency said.

Sarai Sierra, a 33-year-old mother of two, was last heard from on Jan. 21, the day she was due to board her flight back home. Her disappearance attracted a lot of interest in Turkey, where such disappearance of foreign tourists are rare and Istanbul police had set up a special unit to find her.

The Anadolu Agency said the body of a woman was discovered Saturday evening near the remnants of ancient city walls and that police later identified it as Sierra's.

The agency did not say what caused her death. The private NTV television reported that she was stabbed to death, while a private news agency, Dogan, said she had a wound to the head, suggesting she may have been hit by an object.

Police reached by The Associated Press refused to comment on the case.

Sierra, whose children are 9 and 11, had left for Istanbul on Jan. 7 to explore her photography hobby and made a side trip to Amsterdam, Netherlands, and Munich, Germany. She had originally planned to make the trip with a friend, but ended up travelling alone when her friend canceled.

She was in regular contact with friend and family and was last in touch with her family on Jan. 21, the day she was due back in New York. She told them she would visit Galata Bridge, which spans the Golden Horn waterway, to take photos.

The location where the body was found, is a few kilometers away the bridge. It is near a major road that runs alongside the sea of?Marmara and offers an iconic view to visitors of dozens of tankers and other vessels waiting to access the Bosporus strait. Police stopped traffic on the road as forensic police inspected the area.

Anadolu suggested Sierra may have been killed at another location and that her body may have been brought to the site to be hidden amid the city walls.

At least nine people were detained for questioning in Istanbul over the arrests and a police official on the site told journalists two of them were women. The official spoke on condition of anonymity because he was not authorized to speak to reporters on the case.

It was not clear if a Turkish man Sierra had exchanged messages with during her stay in Istanbul was among the group that was detained. That man was detained for questioning on Friday but was later released. Turkish news reports had said Sierra had made arrangements to meet the man on Galata Bridge but the man reportedly told police the meeting never took place.

Sierra's husband, Steven, and brother, David Jimenez, travelled to Istanbul to help in the search. Sierra's mother, Betzaida Jimenez, said she couldn't talk when reached in New York.

Shortly after she was reported missing, Turkey set up a special police unit which scanned through hours of security camera footage in downtown Istanbul in search of clues over her disappearance. A Turkish missing persons association had joined the search, handing out flyers with photos of Sierra and urging anyone with information to call police.

While break-ins and petty thievery is common in Istanbul, the vast and crowded city is?considered relatively safe in comparison to other?major?urban centers. The American's death was unlikely to have a significant impact on tourism, an increasingly large component of the?Turkish economy.

In 2008, an Italian artist Pippa Bacca, was raped and killed while hitchhiking to Israel wearing a wedding dress to plead for peace. Her naked body was found in a forest in northwest Turkey. A Turkish man was sentenced to life in prison for the attack. ?

Detroit's Big Three off to fast sales start in 2013

U.S. auto sales maintained the strong upward trend that delivered a solid send-off to 2012, with most major manufacturers reporting double-digit gains for January.

A number of manufacturers set new sales records during the first month of 2013, and Detroit makers, in particular, recovered some much-needed momentum. Two of the Detroit Big Three ended last year with market share declines.

Industry observers credit a variety of factors for driving strong demand last month, including an improving economy and a bump in the long-struggling housing market. Notably, January saw demand surge in a variety of different segments, from high-mileage small cars and hybrid vehicles to full-size pickups.

?We?re in a fundamentally sound trajectory,? said Mustafa Mohatarem, the chief economist for General Motors. While the industry is still operating well behind the record 17 million-plus rates experienced during the middle of the last decade, he suggested the modest but steady recovery the industry has been experiencing this cycle ?is much more sustainable? than some of the rapid rebounds of decades past.

Though General Motors did see a rise in sales last year, the maker couldn?t keep up with the overall increase in U.S. demand, watching its market share slip from 18.4 percent in 2011 to 16.9 percent. Senior company officials, including CEO Dan Akerson, have forecast a modest improvement for GM this year, and the maker?s 16 percent jump in January appears to put it on an upward trajectory.

Detroit makers, on the whole, appear to have fared well for the month, with Chrysler also gaining 16 percent and marking its 34th consecutive month-over-month sales gains. The maker scored gains at all of its various brands ? and also saw a jump in demand for the Dodge Dart, a critical new compact model that has lagged in showrooms in recent months, something Chrysler CEO Sergio Marchionne has blamed on a lackluster powertrain lineup.

The Detroit Bureau: Handicapping the Super Bowl Auto Ads

Ford, meanwhile, saw January numbers increase by 22 percent. Ken Czubay, the maker?s chief of U.S. sales, suggested that the company's heavy investment in new products and powertrains ?continues to pay off.?

Ford?s results reveal that the January industry upturn was a broad one. Ford?s small car sales jumped 29 percent, while its various hybrid models were up approximately 350 percent. The midsize Ford Fusion had its best January ever. But the full-size F-Series pickup also gained ground.

Pickups have been especially strong in recent months, even as the overall share of small and alternative-powered vehicles has gained steady ground. That?s little surprise, according to analysts such as Joe Phillippi, of AutoTrends Consulting, who track a close, historic link between the housing market and truck sales. Demand for pickups has also gotten a lift as the East Coast continues to dig out from the devastation of Superstorm Sandy.

Detroit makers aren?t the only ones celebrating January?s sales results. Toyota reported a 27 percent year-over-year increase. "The sales pace we saw in the fourth quarter of last year rolled into January, exceeding our expectations for the industry,? said Bill Fay, general manager of the Japanese maker?s mainstream brand.

The other members of the Japanese Big Three didn?t fare quite so well. Nissan gained a modest 2 percent for the month, that increase largely driven by the latest incarnation of the Pathfinder ? which has migrated from a traditional, truck-based platform to a more nimble and fuel-efficient crossover design.

American Honda reported an overall 12.8 percent gain, though its Acura luxury brand did a wee bit better with the assistance of new products such as the entry luxury ILX.

Luxury makers, in general, did well ? but could have done better, some analysts said. Mercedes-Benz gained 11 percent over year-earlier levels, while Audi was up nearly 6 percent -- but both reported records for January.

The Detroit Bureau: Honda to Integrate Apple Siri Personal Assistant

BMW was up a modest 2.3 percent after finishing 2012 as the nation?s best-selling luxury brand. Some observers suggest the maker?s aggressive end-of-year marketing may have ?pulled forward? sales that might have otherwise been rung up in 2013.

But Jesse Toprak, auto analyst with data tracking firm TrueCar.com, also said that the new tax increases on wealthy buyers may be having a measurable impact on high-line brands going forward.

TrueCar data indicate that buyers actually paid a slight bit less for the average vehicle sold in January, at $30,812, compared with December. But the average transaction price was nonetheless up by $416, or more than 1percent year-over-year.

Meanwhile, with demand helping makers maintain steady production schedules on most key products, they?ve been able to continue a year-long reduction in rebates and other incentives. For the month, such givebacks slid to just $2,274 per vehicle, down from $2,591 in December and $2,481 in January 2012.

The Detroit Bureau: Chrysler Planning 47 New Models Over Next Four Years

Only a few automakers increased givebacks, notably Hyundai, which saw a weak 2 percent gain in sales in January. Like several other makers, however, the Korean brand has warned it could face production-driven limits to its sales if the market continues to heat up too quickly.

Earlier this week, AutoPacific, Inc. predicted the U.S. market would climb to 15.1 million this year following last year?s jump to 14.5 million. But the California-based consulting firm is among the more conservative. Chrysler officials indicated they see a 15.5 million total for 2013 if current trends hold.

Whether they can, of course, is far from certain. The U.S. economy is clearly gaining momentum but there are a variety of ?headwinds,? industry insiders caution. That includes the ongoing rise in petroleum prices ? which could climb even faster in the wake of the explosion at Mexico?s national oil company headquarters and the terrorist attack on the U.S. embassy in Turkey this week.

There?s also the ongoing drag from the European economy. Car sales there fell to their lowest level since 1995 last year and few expect any improvement in 2013. With China also slowing, officials with foreign manufacturers, such as Toyota?s Fay, have said they will be putting even more emphasis on the North American market in the months ahead.

After the sharp cutbacks of the recent recession, the auto industry could translate improved cost-efficiencies into record profits this year ? but should makers decide to battle it out by ramping up incentives, analysts warn, that could stall the ongoing recovery at the bottom line level.

Genome-wide atlas of gene enhancers in the brain online

Friday, February 1, 2013

Future research into the underlying causes of neurological disorders such as autism, epilepsy and schizophrenia, should greatly benefit from a first-of-its-kind atlas of gene-enhancers in the cerebrum (telencephalon). This new atlas, developed by a team led by researchers with the U.S. Department of Energy (DOE)'s Lawrence Berkeley National Laboratory (Berkeley Lab) is a publicly accessible Web-based collection of data that identifies and locates thousands of gene-regulating elements in a region of the brain that is of critical importance for cognition, motor functions and emotion.

"Understanding how the brain develops and functions, and how it malfunctions in neurological disorders, remains one of the most daunting challenges in contemporary science," says Axel Visel, a geneticist with Berkeley Lab's Genomics Division. "We've created a genome-wide digital atlas of gene enhancers in the human brain - the switches that tell genes when and where they need to be switched on or off. This enhancer atlas will enable other scientists to study in more detail how individual genes are regulated during development of the brain, and how genetic mutations may impact human neurological disorders."

Visel is the corresponding author of a paper in the journal Cell that describes this work. The paper is titled "A High-Resolution Enhancer Atlas of the Developing Telencephalon." (See below for a list of co-authors.)

The cerebrum is the most highly developed region of the human brain. It houses the cerebral cortex, the so-called "gray matter" where complex information processing events take place, and the basal ganglia, a brain region that helps control movement throughout the body and is involved in certain types of learning. Many of the genes responsible for development of the cerebrum have been identified but most of the DNA elements responsible for expressing these genes - turning them on/off - have not. This is especially true for gene enhancers, sequences of DNA that act to amplify the expression of a specific gene. Characterizing gene enhancers tends to be difficult because an enhancer does not have to be located directly adjacent to the gene it is enhancing, but can in fact be located hundreds of thousands of DNA basepairs away.

"In addition to acting over long distances and being located upstream, downstream or in introns of protein-coding genes, the sequence features of gene enhancers are poorly understood," Visel says. "However, gene-centric studies have provided strong evidence that gene enhancers are critical for normal embryonic development of the brain and have also linked human diseases to perturbed enhancer sequences."

Visel and an international team of researchers met the challenges of systematic identification and functional characterization of gene enhancers in the cerebrum through a combination of ChIP-seq studies and large-scale histological analyses in transgenic mice, in which the activity patterns of human telencephalon enhancers can be studied. ChIP-seq, which stands for "chromatin immunoprecipitation followed by sequencing," is a technique for genome-wide profiling of proteins that interact with DNA.

This combination of approaches enabled Visel and his colleagues to identify over 4,600 candidate embryonic forebrain enhancers. Furthermore, studying mouse embryos they mapped the activity of 145 of these enhancers at high resolution to define where exactly in the developing brain they drive the expression of their respective target genes. The result is a comprehensive, electronically accessible database for investigating the gene regulatory mechanisms of cerebrum development, and for studying the roles of distant-acting enhancers in neurodevelopmental disorders.

"By mapping hundreds of gene enhancer sequences and defining where exactly in the developing brain they are active, our enhancer atlas provides important information to connect non-coding mutations to actual biological functions," Visel says.

As an example of how the enhancer atlas could be used in this manner, Visel says that if through genetic studies a certain region of the human genome has been linked to a specific neurological disorder but that region does not contain any protein-coding genes, checking the atlas could reveal whether the region is home to any distant-acting gene enhancers.

"If there is a gene enhancer in the region," Visel says, "researchers could determine exactly how the enhancer or a mutated form is related to the disorder. This encompasses a lot of territory in the human genome. More than half of all the disease-related DNA sequence changes discovered in genome-wide screens to date fall within the 98-percent of human genome sequences that do not code for proteins. These 'non-coding' sequences were once dismissed by many as 'junk DNA' but are now known to harbor critical gene regulatory functions."

Visel says the enhancer atlas also provides a "molecular toolbox" that can be used to re-locate the expression of certain genes to specific regions of the brain in experimental applications. This could help researchers gain new insights into how the brain develops and how the human brain has evolved.

"The evolution of the gene regulatory architecture may have been one of the drivers of the extremely advanced brains that we see in primates in general and humans in particular," he says.

While continuing to map and annotate additional enhancers, Visel and his colleagues are now also studying the biological mechanisms related to some of these enhancers in more detail.

"We still only partially understand how gene enhancers work in the context of a living brain, i.e., what proteins bind to them, how mutations change their function, and what exactly the role of these sequences is in the context of human neurological disorders," Visel says. "Clearly, there is more work to do."

Thousands of Vukovar Croats rally against Serb Cyrillic signs

VUKOVAR, Croatia (Reuters) - More than 20,000 Croats rallied on Saturday in Vukovar, a town destroyed in the 1991-95 war with rebel Serbs, to protest against the government's plan to introduce signs in the Serbian Cyrillic alphabet.

The Social Democrat-led government, which will take the country into the European Union on July 1, wants to put up bilingual signs, in both the Latin script used for Croatian and in Cyrillic, in areas where the population is more than one third ethnic Serb later this month.

Croatia already has bilingual signs, in Croatian and Italian, in the northern Adriatic Istrian peninsula, close to Italy, but the issue of Serb language and script is much more sensitive in the country still traumatized by the war.

According to the 2011 census, there are about a dozen regions in Croatia with a sizeable Serb community, including the easternmost town of Vukovar, which many Croats still see as a symbol of destruction and suffering brought on by the Serb rebellion against Croatia's independence from Yugoslavia.

The protest was organized by war veteran groups under the slogan "For a Croatian Vukovar, No to Cyrillic". The protesters waved Croatian flags and banners reading "This is not what we fought for" and "Don't test our patience".

Tomislav Josic, one of the organizers, said they wanted the law on minority rights, which stipulates bilingual signs, not to be applied to Vukovar "as a sign of respect for the sacrifice Vukovar has made".

The picturesque town on the Danube was reduced to rubble during a three-month siege by the Serb-dominated Yugoslav army and local Serb militia in 1991. All non-Serbs were expelled and some 200 patients from a local hospital were executed after its capture by the Serbs.

It has been rebuilt, but the area remains poor, with high unemployment and lingering ethnic tensions.

"For all of us who suffered in Vukovar, it is difficult to accept (Cyrillic script). It is too early for that, after all the victims, all the suffering we've been through," said Nada Soldi, a Vukovar pensioner who attended the rally.

The Croatian and Serbian languages are mutually intelligible, but Croats, who tend to be Roman Catholic, use the Latin script, while traditionally Orthodox Serbs use a Cyrillic alphabet similar to that of Russian.

The Sinister Scribblings of Sarah E. Glenn: Interview: Lillian Brummett

Today, I welcome Lillian Brummet to Sinister Scribblings. Lillian is an author, but also so much more. She and her husband Dave offer book publishing services when they're not promoting green living and living positively. They also host an Internet Radio program, "World of Writing", on her channel, Conscious Discussions Talk Radio:
The next broadcasts are scheduled for February 7th and 17th.

Today, she's answering some questions I posed to her about self-promotion.

Q: Whenever I read blogs about promoting writing, I keep hearing about a 'platform'. What do they mean by this term? Is it the same thing as the buzzword 'branding yourself'?

You are so right about the changing buzzwords in the industry ? people are coming up with new terms to mean similar things and it can be confusing that is for sure. So I?ll try to clear this up for you and your readers.?

Branding: this term is used to describe how you present yourself or better said, how you appear to others, or what you are known by. An example of this would be: ?Jane has have built a reputation for being a green living expert that specializes on educating her audience. So that is her brand. This also means that her logo, the colors she uses in her promotion materials, the website? everything that her put out there reflects this brand.?

Platform: this term is used to describe how you get your branding known. It is kind of like a launching pad that you built that supports your marketing and promotion efforts. So this would include promotion materials you?ve prepared, a website that describes your services and/or products, blog, setting up social media, newsletter, e-newsletter, radio show, video series? or whatever you are choosing to utilize. When you have a platform created (and this can be an ongoing evolving process) it offers you a solid place to stand on and makes your other efforts more efficient.?

Q: I had a self-published author tell me once that you needed to have 5 books in print to make enough money to support yourself. Is this true, presuming that one lives modestly??

Honestly the comment you heard does make sense in that book sales can fluctuate and the more books you have the better chance you have of creating a sustainable steady income. Sadly, though, in reality this?isn't?necessarily the case. For instance?I've?met authors who have written a small book, one single book, but then created a steady income out of it. They?ve built speaking engagements, alternate formats of the book (think audio CD?s, streaming video series, webinars, etc.) and have strong marketing skills. Other authors may have written 10 books and still have to work that ?day? job in order to make ends meet. So I would say that income is really an individual thing ?your book last year might do well but next year?s book flops for the first year, but then sales pick up again 3 years later.?

I think you just have to have faith in yourself and diversify so that you can have a steady stream of income (such as doing freelance work or offering workshops at the local college). The key to dealing with the low moments is to remind yourself why you are writing in the first place. If you are doing this to leave a legacy, because you love the craft of writing, because this allows you a chance to create positive change, to help evolve society, or some other greater good ? then you can console yourself, knowing that this is more important than fiscal gain.

Q: Is it worth the money to buy ads on Facebook, Goodreads, or similar sites??

I am very wary of putting out money for advertising as an author. I?ve paid for ads early on in my career in both print and online publications/sites and haven?t had a good Return On Investment (ROI). After becoming a staff writer and member of the media, I learned that there are a lot of ways of saving money and creating a better ROI, which I share in our Purple Snowflake Marketing (PSM) book. I have found that there is a higher ROI for ads highlighting either the radio show or blog, and then from there they learn about our books and services. Sites like what you are talking about often offer what is called Pay Per Click (PPC) and this simply means that you don?t pay anything for your ad until someone clicks on it ? and you might pay a few cents per click. But that can be dangerous to the budget if all of a sudden you have 5,000 clicks! So you might want to set up a limit if you go this route. The trick with advertising is to understand what your agenda is for that ad ? meaning, do you want to drive traffic to your blog or website, or sell a specific product. Once you know what your agenda is for that one ad, you can then create engaging content for the ad that inspires the reader to what is deemed a: Call To Action (CTA). However, once you?ve engaged the reader to respond to your CTA is only the first step ? what they see when the go to where you sent them is also an important thing to focus on. So, say for instance you?ve decided to send people who read your ad to your website home page ? what will they see there and how does that help build your branding or increase your income?

Q: Could you offer us one tip on generating sales after the Honeymoon Period (described in Purple Snowflake Marketing)?

Ah? this is a great question. First, let me define the Honeymoon Period for your readers ? this is the first year after a book is released, when the author has exhausted current contact lists and local outlets, and the author has usually run out of their initial promotional budget. The author?s excitement level also tends to run lower after the first year and they begin to tire of marketing the same product over and over.?

So my tip today would be to get creative ? I mean, yes you do want to do all those activities in the first year, but the life of your book may be 7 years or longer depending on the contract with your publisher. Branch out to other communities that surround your area, find new ways of getting media exposure, look for long-term promotion materials (like bookmarks) and get involved with organizations, and get busy online where there are tens of thousands of opportunities just waiting for you.

Q: Can an introvert successfully promote and publicize her writing? Never mind why I'm asking this.

(She laughs) I love that you ask this, since I am an introvert myself. The thing is that each of us has unique strengths and weaknesses, we have a unique location, a unique budget for each and every book we release. We need to constantly evaluate where we are at and what we can do. You?ll notice that our PSM book talks about creating a business plan for your career as an author, but then a marketing plan for each book you release. Those plans should be very flexible and should probably be revised every 3-6 months. Who you are on a personal level plays a vital role in this.?

Most of us will try different things and go through a bit of a learning curve before we figure out what works best for us. Some of us are great at one on one networking, others do very well at speaking events; someone might be a great teacher, while others have fantastic technical skills. One person might excel at radio interviews, another might find better results through social networking, and someone else finds out that they have great results through blog appearances, or promotional articles in a variety of publications. Everyone is different and I think this is a good thing ? after all we want to stand out and be noticed? not do what everyone else is doing and get lost in the crowd.?

Thank you for spending time with us. I appreciate it. Below is information on Purple Snowflake Marketing, for authors who would like to learn more about promoting their work.
